Background
The 33A10-defined antigen is the orphan protein, Placenta expressed transcript (Plet)-1. Biochemical data indiCated that Plet-1 is a glycosylphosphatidylinositol-anchored glycoprotein with N-linked carbohydRates in addition to other posttranslational modifiCations. Plet-1 is a a recently identified protein that as of yet has no known function and is not associated with any one family of proteins. Plet-1 is used as a cell surface marker present on stem/progenitor epithelial cells and in a number of tissues including thymus and skin, and on epithelial cell types in mammary gland, prostate, and pancreas.
Source
33A10 is a Rat IgG2b monoclonal antibody derived by fusion of SP2/0 Mouse myeloma cells with spleen cells of Sprague-Dawley Rats immunized with Mouse mammary tumor cells.
Product
Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
Formulation: Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
Specificity
33A10 reactivity is identified in the developing Mouse mammary gland, in the bronchioles of the lung, the salivary glands and the skin. Plet-1 is a product specific for Keratinocytes undergoing terminal trichilemmal differentiation in the hair follicle and is expressed at the plasma membranes.
Applications
33A10 is useful for Flow Cytometry, Immunofluorescence, Immunohistochemistry (frozen). Optimal antibody dilution should be determined by titration.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
