Background
The reactivity of the antiserum is restricted to the Fc part of the IgG molecule. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against normal monkey serum a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgG. No precipitation reaction is obtained with purified IgA, IgM, and IgG/Fab fragments. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgG in monkey serum or other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of monkey IgG from serum or plasma.
Source
Highly purified normal IgG isolated from pooled rhesus monkey serum. Freund’s complete adjuvant is used in the first step of the immunization procedure
Product
Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.
Formulation: Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added.
Specificity
Precipitating polyclonal Rabbit antiSerum to Monkey IgG, Fc specific
Species Reactivity: The antiSerum does not cross react with any other component of the Monkey immunoglobulin system. Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Precipitation reactions have been observed with IgG in Serum of other old-world Monkeys, including Cercopithecus, Cynomolgus and baboon. The antiSerum may also react with IgG of other species as has been observed for Chimpanzee and man.
Applications
Precipitation assays. In immunoelectrophoresis use 2 μl serum or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in 3 mm diameter center well and 2 μl serum samples (neat and serially diluted in 2 mm diameter peripheral wells
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
