Background
Mucin 1, cell surface associated (MUC-1) or polymorphic epithelial mucin (PEM) is a mucin encoded by the MUC1 gene in Humans. MUC-1 is a glycoprotein with extensive O-linked glycosylation of its extracellular domain. Mucins line the apical surface of epithelial cells in the lungs, stomach, intestines, eyes and several other organs. The protein serves a protective function by binding to pathogens and also functions in a cell signaling capacity. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with colon, breast, ovarian, lung and pancreatic cancers.
Product
Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
Formulation: Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
Specificity
175C5 shows a high preference for breast carcinomas relative to normal breast epithelium. The antibdy reacts with a carcinoma-associated antigen in both adenocarcinomas and squamous cell carcinomas of different origins.
Applications
140 H2 is useful for ELISA, immunofluorescence, flow cytometry, immunoprecipitation, western blot, electron microscopy and immunohistochemistry on frozen tissues. Optimal antibody dilution should be determined by titration.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
