Background
Members of the tumor necrosis factor receptor (TNFR) family are emerging as important molecules implicated in the regulation of proliferation, differentiation and survival of T and B lymphocytes. Among these receptors is CD27, the function of which has thus far only been studied in the human system, where it amplifies the T cell proliferative response induced by TCR triggering. The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is required for generation and long-term maintenance of T cell immunity. It binds to ligand CD70, and plays a key role in regulating B-cell activation and immunoglobulin synthesis. This receptor transduces signals that lead to the activation of NF-κB and MAPK8/JNK. Adaptor proteins TRAF2 and TRAF5 have been shown to mediate the signaling process of this receptor. CD27-binding protein (SIVA), a pro-apoptotic protein, can bind to this receptor and is thought to play an important role in the apoptosis induced by this receptor.

mAbs to murine CD27 were generated injecting a mixture of the mCD27tag expresssing clones AR0524 and AR0530; Hamster spleen cells and SP2/0 murine myeloma cells were fused.

Specificity
LG.3A10 and LG.8A6 see a different epitope. A combination can therefore be developed into a double determinant ELISA to detect soluble Mouse CD27. LG.3A10 cross-react with Human CD27, LG.8A6 does not.
Applications
LG.3A10 is useful for flow cytometry, immunoprecipitation and immunohistochemistry on frozen and paraffin embedded sections . Optimal antibody dilution should be determined by titration. Recommended dilution for immunohistochemistry: 1:100. In combination with LG.8A6, it is possible to develop a double determinant ELISA to detect soluble Mouse CD27.
Storage
The antibody may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
